摘要

目的:探究石榴籽油(pomegranate seed oil, PSO)和鱼油(fish oil, FO)对膝骨性关节炎(knee osteoarthritis, KOA)大鼠膝关节软骨IL-1βmRNA表达的影响因素,比较石榴籽油和鱼油对膝关节性关节炎的作用关系,为KOA防治拓展新的思路并提供科学依据。方法:选取8周健康雌性SD大鼠154只,将实验动物用EXCEL随机分成7组:假手术(Sham)组、模型(Model)组、生理盐水(NS)组、石榴籽油低剂量(PSOLD)组、石榴籽油高剂量(PSOHD)组、鱼油低剂量(FOLD)组、鱼油高剂量(FOHD)组,每组22只。采用改良Hulth法手术建立大鼠KOA模型,选取有代表性的两种ω-3多不饱和脂肪酸(n-3PUFAs)PSO和FO实验组干预,Sham组与Model组不做干预,NS组大鼠每天使用生理盐水1 mL/kg灌胃,PSOLD组、PSOHD组分别给予0.83 mL/kg、1.66 mL/kg剂量PSO灌胃,FOLD组、FOHD组分别予0.69 mL/kg、1.38 mL/kg剂量FO灌胃。干预8周后,每组随机处死10只,取大鼠后肢膝关节软骨标本,进行RT-PCR;干预 12周后,处死各组剩余10只大鼠,处理方式同上。结果:与Sham组比,Model组及各试验组8周、12周IL-1βmRNA相对表达量显著升高(P<0.01);与Model组相比,PSO和FO组8周、12周IL-1βmRNA相对表达量均显著降低(P<0.01);其余各组差异无统计学意义(P>0.05)。Model组和NS组8周与12周IL-1βmRNA相对表达量对比有显著增加(P<0.05),其余各组差异无统计学意义(P>0.05)。结论:(1)石榴籽油和鱼油都可以有效降低IL-1βmRNA相对表达量,使软骨退变得到改善;(2)相同干预时长的PSOLD组和PSOHD组,FOLD组和FOHD组,下调IL-1βmRNA表达水平的作用均无显著性差别;(3)从8周和12周数据分析对比,两组PSO组和FO组对IL-1βmRNA表达量与干预时长相关性不显著。

Abstract

Objective: To explore the influencing factors of pomegranate seed oil and fish oil on the expression of IL-1βmRNA in knee cartilage in rats with osteoarthritis (OA), and to compare the effects of pomegranate seed oil and fish oil on knee arthritis (KOA), so as to expand new ideas and provide scientific basis for the prevention and treatment of KOA. Methods: A total of 154 8-week-old healthy female SD rats were selected, and the experimental animals were randomly divided into 7 groups by EXCEL: Sham group, Model group, normal saline (NS) group, pomegranate seed oil low-dose (PSOLD) group, pomegranate seed oil high-dose (PSOHD) group, fish oil low-dose (FOLD) group, and fish oil high-dose (FOHD) group, with 22 rats in each group. The modified Hulth method was used to establish the rat KOA model, and the representative two omega-3 polyunsaturated fatty acids (n-3PUFAs), pomegranate seed oil (PSO) and fish oil (FO) experimental groups were selected for intervention, and the Sham group and the Model group did not intervene, the rats in the NS group were given 1 ml/kg of normal saline per day, and the PSOLD group and PSOHD group were given 0.83 ml/kg and 1.66 ml/kg PSO gavage, respectively. The FOHD group was given 0.69 ml/kg and 1.38 ml/kg by gavage, respectively. After 8 weeks of intervention, 10 rats in each group were randomly sacrificed, and the cartilage specimens of the knee joint of the hind limbs of the rats were taken for RT-PCR. After 12 weeks of intervention, the remaining 10 rats in each group were sacrificed, and the treatment was the same as above. Results: Compared with the Sham group, the relative expression of IL-1β mRNA in the Model group and the experimental groups was significantly increased at 8 weeks and 12 weeks (P<0.01). Compared with the Model group, the relative expression of IL-1β mRNA in the PSO and FO groups was significantly decreased at 8 and 12 weeks (P<0.01). There was no significant difference among the other groups (P>0.05). The relative expression of IL-1β mRNA in Model group and NS group increased significantly at 8 weeks and 12 weeks (P<0.05), while there was no significant difference between the other groups (P>0.05). Conclusion: (1) Pomegranate seed oil and fish oil can effectively reduce the relative expression of IL-1βmRNA and improve cartilage regression. (2) There is no significant difference in the effect of down-regulating IL-1βmRNA expression between the PSOLD group and the PSOHD group, the FOLD group and the FOHD group with the same intervention duration. (3) From the analysis of 8-week and 12-week data, there is no significant correlation between the expression of IL-1β mRNA and the length of intervention in the two groups of PSO and FO groups.
