紫杉醇在自体动静脉内瘘局部血管投递部位病理变化的研究*

摘要

目的: 探讨紫杉醇在自体动静脉内瘘局部血管投递的短期效果及可能机制。方法: 将大耳白兔36只随机分为观察组和对照组,动物模型制作完成后在2周、4周、6周、8周、10周、12周分别手术取出动静脉内瘘血管,通过HE染色观察静脉血管组织形态学变化,通过免疫组化法观察静脉血管壁Beclin-1、LC3Ⅱ及PDGF-bb因子表达情况。结果: 观察组静脉内膜厚度低于对照组(P<0.05),Beclin-1、LC3Ⅱ及PDGF-bb因子表达观察组略低于对照组,差异无统计学意义(P>0.05)。结论: 自体动静脉内瘘局部血管投递紫杉醇可降低静脉内膜异常增生,为自体动静脉内瘘(AVF)狭窄提供了新的治疗思路。

Abstract

Objective: To investigate the short-term effect and possible mechanism of paclitaxel in local vascular delivery of autogenous arteriovenous fistula(AVF). Methods: Thirty-six white rabbits were randomly divided into observation group and control group. After the completion of the animal model, AVF vessels in each group were surgically removed at 2 weeks, 4 weeks, 6 weeks, 8 weeks, 10 weeks and 12 weeks, and the morphological changes of venous vessels were observed by HE staining. The expressions of Beclin-1, LC3Ⅱ and PDGF-bb in venous wall were observed by immunohistochemistry. Results: The intima venous thickness in the observation group was lower than that in the control group (P<0.05), and the expression of Beclin-1, LC3Ⅱ and PDGF-bb in the observation group was slightly lower than that in the control group, with no statistical significance (P>0.05). Conclusion: Local delivery of paclitaxel to autologous AVF can reduce the abnormal hyperplasia of the venous intima. It provides a new therapeutic idea for AVF stenosis.
