Objective: To investigated the expression of receptor-interacting protein kinase 3 (RIPK3) in gastric adenocarcinoma and precancerous tissues. Methods: The study focuses on 170 pathological samples of gastric adenocarcinoma and precancerous lesions collected from the Second Affiliated Hospital of Baotou Medical College from December 2022 to June 2024, pathological samples including 40 cases of chronic superficial gastritis, 40 cases of chronic atrophic gastritis with intestinal metaplasia, 40 cases of heterotrophic hyperplasia and 50 cases of gastric adenocarcinoma. To use immunohistochemistry to detect the expression of RIPK3. Meanwhile, 30 fresh tissue samples are collected to detect the expression of RIPK3 using Western blot. The relationship between RIPK3 expression and survival of gastric adenocarcinoma patients is analysed by Kaplan-Meier plotter, and the association between RIPK3 and the tumour microenvironment (TME) of gastric cancer is explored using TISCH database. Results: (1) Immunohistochemistry shows that the positive expression rate of RIPK3 in gastric adenocarcinoma is significantly lower than that in other groups (P<0.05) and correlated with the depth of tumour infiltration, TNM stage, vascular invasion and lymph node metastasis (P<0.05). (2) Western blot results shows that the expression of RIPK3 in chronic superficial gastritis, chronic atrophic gastritis with intestinal metaplasia and gastric adenocarcinoma gradually decreased (P<0.01). (3) Kaplan-Meier analysis shows that gastric adenocarcinoma patients with high RIPK3 expression has longer overall survival (OS) and progression-free survival (FP). (4) TME analysis reveals that RIPK3 expression is closely associated with intercellular stroma and mucus secretion. Conclusion: Low expression of RIPK3 is associated with gastric adenocarcinoma occurrence, infiltration, metastasis and poor prognosis.
