Expression level of hypoxia inducible factor-1 Cationits clinical significance to sjögren's syndrome

Abstract

Objective: To analyze the correlation between the expression level of hypoxia inducible factor-1α (HIF-1α)and relevant laboratory indicators and dry mouth symptoms in Sjögren's syndrome(SS) patients by comparing the expression levels of HIF-1α in peripheral blood between healthy individuals and patients with SS.Methods: A total of 64 SS patients and 64 health examiners were selected. PCR was used to detect the expression level of HIF-1α mRNA in peripheral blood mononuclear cells (PBMC) of the two groups. ELISA method was used to detect the expression level of HIF-1α protein in serum. Immunoglobulin (Ig), erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), white blood cell (WBC), hemoglobin (Hb) and other indicators from SS patients were collected. Results: Compared with the healthy control group, the expression levels of HIF-1α mRNA in PBMC were increased (P<0.05). HIF-1α expression level in serum of SS patients was positively correlated with ESR, IgG and IgM (r1=0.457 6, r2=0.426 9, r3=0.403 9, P<0.05), but negatively correlated with WBC and Hb(r1=-0.333 3, r2=-0.312 5, P<0.05. The expression level of HIF-1 protein in positive dry mouth patients was higher than that in negative dry mouth patients (P<0.01). The diagnostic value of the expression level of HIF-1α mRNA and protein on SS was analyzed using ROC curves, and the AUC was 0.659 4 and 0.813 4, with the sensitivity of 64.10% and 76.60%, and the specificity of 95.30% and 81.20%, respectively. Conclusion: The expression level of HIF-1α mRNA and protein were higher in PBMC of SS patients. It is positively correlated with ESR, IgG and IgM, while negatively correlated with WBC and Hb. The expression level of HIF-1α in SS patients with positive dry mouth was higher than that of SS patients with negative dry mouth, indicating that HIF-1α has certain diagnostic value in SS and plays a role in the occurrence of SS.
