Protective effect of APX-115 free base on H9c2 cells against high concentration of glucose

Abstract

Objective: To investigate the protective effect of Nox inhibitor APX-115 free base on H9c2 cells against high concentration of glucose. Methods: The proliferative activity of H9c2 cells after pretreatment with different concentrations of APX-115 free base was detected by CCK8 assay. The expression of Nox4 protein was detected by Western blot and immunofluorescence. The morphological changes of H9c2 cells were observed. Results: Compared with the Model group, whrn the concentration of APX-115 free base was 8 μmol/L, the cell proliferation activity decreased, and the cell proliferation activity increased at other concentrations. The expression of Nox4 protein in each treatment group of APX-115 free base was down-regulated. The cell morphology of the APX-115 free base 5 μmol/L treatment group returned to spindle shape, the cell distribution was gradually uniform, the intercellular space was reduced, and the number of cells basically returned to normal level. Conclusion: When APX-115 free base is 5 μmol/L, it has the best protective effect on H9c2 cells induced by high glucose.
