Effect of Nuangong Qiwei Pill on ovarian function in mice with    premature ovarian failure induced by cyclophosphamide

Abstract

Objective: To observe the protective effect of Mongolian medicine Nuangong Qiwei Pill on cyclophosphamide-induced premature ovarian failure mice. Methods: Female mice with normal estrous cycle were randomly divided into 5 groups : blank control group, cyclophosphamide group, low dose group, medium dose group and high dose group of Nuangong Qiwei Pill. Except for the blank control group, the mice in the other groups were intraperitoneally injected with cyclophosphamide to prepare the premature ovarian failure model (the first dose was 50 mg/kg, followed by 8 mg/kg/d for 14 days). After the modeling, the blank control group and the cyclophosphamide group were given 0.1 ml normal saline by gavage, and the low, medium and high dose groups of Nuangong Qiwei Pill were given 0.25, 0.5 and 0.8 g/kg by gavage for 14 days. The changes of body weight and estrous cycle of mice in each group were monitored daily. After the intervention, the serum of mice was collected to detect the levels of anti-mullerian hormone (AMH), estradiol (E2) and follicle stimulating hormone (FSH), and the ovarian tissue sections were stained and analyzed. Results: Compared with the blank control group, the body weight of the mice in the cyclophosphamide group decreased, the estrous cycle disappeared, the serum E2 and AMH levels decreased significantly, and the FSH level increased significantly (P<0.01). Ovarian sections showed a significant increase in atretic follicles and interstitial fibrosis. Compared with the cyclophosphamide group, there was no significant difference in body weight of mice in each dose group of Nuangong Qiwei Pills; the estrous cycle of mice in the medium and high dose groups was significantly restored; the serum E2 level of mice in the high dose group was significantly increased (P<0.05), the AMH and FSH levels did not change significantly (P>0.05), and the ovarian tissue and structure were improved to a certain extent. Conclusion: Mongolian medicine Nuangong Qiwei Pill can improve the ovarian function of cyclophosphamide-induced model to a certain extent. High dose treatment for 14 days can significantly restore the estrous cycle, improve serum hormone levels, repair damaged ovarian tissues, and play a certain therapeutic role in premature ovarian failure.
