Effect of hypoxic preconditioning on NK cell killing activity in mice

Abstract

Objective:To investigate the effect of hypoxic preconditioning (HPC) on NK cell killing activity in mice. Methods: Selected mice were divided into the control group (Group A), acute hypoxic group (Group B) and HPC group (Group C). The levels of interferon-γsecreted by NK cells were detected with EILSA, expression levels of perforin and granzyme were detected with Real-Time PCR, and the killing activities of NK cells of mice in the three groups were detected by MTT colorimetry. SPSS 19.0 statistical software was used for data analysis. Results: Compared with the group B, the killing activity of HPC group (Group C) was more powerful, and it obviously enhanced at 12 h(P<0.05). The secretion of IFN-γ by NK cells was gradually decreased from 4 h to 16 h, which indicated that hypoxia preconditioning was more adaptive to hypoxia than the group B over time. Compared with the group B, the hypoxic injury of mice in the HPC group were relieved, and expression levels of perforin and granzyme were higher than that in the group B(P<0.05). Conclusion: Compared with the acute hypoxic group, NK cell killing function in the HPC group was recovered, and secretions of IFN-γ, perforin and granzyme protein were increased.
