Objective: To investigate the application value of interleukin (IL)-8 and E-cadherin (E-cad) in the diagnosis and prognosis of cervical cancer. Methods: (1)A total of 100 patients with cervical cancer who were admitted to the hospital from January 2020 to December 2021 were collected, and 100 of their cancer tissues were taken as the observation group, and 100 of their adjacent tissues were taken as the control group. The expression of IL-8 and E-cad in the two groups was determined by real-time fluorescence quantitative polymerase chain reaction (PCR). The expression of IL-8 and E-cad in cervical cancer patients with different clinical parameters was compared. (2)RT-qPCR was used to determine the expression of serum IL-8 and E-cad in 100 patients with cervical cancer and 100 healthy people in the same period. Receiver operating characteristic curve (ROC) was used to analyze the diagnostic value of combined detection of IL-8 and E-cad in cervical cancer. (3)CCK8 and WB were used to detect the changes of proliferation and apoptosis in human cervical cancer cell line HeLa groups (blank control, IL-8 inhibitor, E-cad inhibitor and IL-8/E-cad inhibitor), and to study its application value in the progression of cervical cancer. Results: (1)The levels of IL-8 and E-cad in the observation group were higher than those in the control group (all P<0.05). Comparing the expression of IL-8 and E-cad in different clinical parameters of cervical cancer patients, it was found that there were significant differences in tumor diameter, TNM stage, organ and lymph node metastasis (all P<0.05). (2)When cervical cancer was diagnosed alone, the area under the curve (AUC) of IL-8 was 0.741, and the 95% confidence interval was (0.642,0.839); the area under the curve (AUC) of E-cad was 0.757, and the 95% confidence interval was (0.658, 0.855). When the two were combined to diagnose cervical cancer, the area under the curve (AUC) was 0.838, and the 95% confidence interval was (0.756, 0.919), the difference was statistically significant (P<0.05). (3)At 24 h, 48 h and 72 h, the absorbance of IL-8 inhibitor group, E-cad inhibitor group and IL-8/E-cad inhibitor group was lower than that of blank control group, which inhibited proliferation, and the difference was statistically significant (P<0.05). The expression of Cleaved-Caspase3/Caspase3 in IL-8 inhibitor group, E-cad inhibitor group and IL-8/E-cad inhibitor group was higher than that in blank control group, which promoted apoptosis, and the difference was statistically significant (all P<0.05). Conclusion: Combined detection of IL-8 and E-cad can significantly improve the diagnostic value of cervical cancer compared with single detection, and has a certain monitoring effect on its prognosis.
