Objective: To investigate the effect of resveratrol (RSV) on renal function in high-fat mice and its mechanism of regulating SIRT1 to inhibit oxidative stress and improving renal injury in mice. Methods: Thirty six-week-old C57BL/6J mice were selected to construct a high-fat mouse model. The mice were divided into the control group (CTRL group), the high-fat group (HL group), and the resveratrol group (RSV group), with 10 mice in each group. After 18 weeks of feeding, the blood was collected to detect the levels of blood lipid and creatinine (CRE). The pathological changes of kidney were observed by HE and immunohistochemical staining. The changes of SIRT1 and oxidative stress related proteins in mice kidney were observed by Western blot and RT-PCR. Results: The levels of blood lipid TG, TC, LDL-C and creatinine (CRE) in RSV group were decreased, the mesangial proliferation and fibrosis in renal tissue were decreased, the expression of fibronectin (FN) was increased, and the expression of SIRT1 and antioxidant enzymes GPx4 and SOD2 in mice kidney were increased. Conclusion: RSV improves the expression of antioxidant enzymes by regulating the SIRT1 pathway, and inhibits oxidative stress in renal tissue to improve renal function injury induced by high-fat mice.
