Objective: To study the effect of storage in vitro on Eryptosis. Methods: Recruit 6 healthy adult volunteer blood donors, collect 400 mL of anticoagulated whole blood, filter and remove the plasma by centrifugation, resuspend red blood cells (RBC) in 100 mL MAP red blood cell protection solution, and store at 4 ℃ . 6 mL of blood samples were drawn at the 0, 7, 14, 21, 28, and 35th day of the storage period respectively. The RBC membrane Phosphatidylserine (PS) content was measured by flow cytometry, automatic blood cell counter, blood gas analyzer, etc, mean corpuscular volume (MCV), extracellular ion concentration, extracellular glucose (Glucose, Glu), lactic acid (Lactic acid, Lac), total bilirubin (Total bilirubin, TBil) concentration and methemoglobin fraction (Fraction of Methemoglobin, FMetHb). Results: With the prolonged storage time, PS content decreased (r=-0.8356), MCV, extracellular Cl- concentration did not change, extracellular TBi concentration (r=0.8035) ,Lac concentration (r=0.9377) and FMetHb (r=0.9706) .The concentration of Na+ (r=-0.9274) and Glu (r=-0.9860) in the cells decreased with the extension of storage time. Conclusion: In vitro storage does not cause increased RBC death.
