目的: 探究白藜芦醇(RSV)对高脂小鼠肾功能的影响及其调控SIRT1抑制氧化应激反应改善小鼠肾损伤的作用机制。方法: 选取30只6周龄C57BL/6J小鼠构建高脂小鼠模型,将小鼠分为对照组(CTRL组)、高脂组(HL组)、白藜芦醇组(RSV组)三组,各10只。喂养18周后取血检测小鼠血脂、肌酐(CRE)水平;采用HE、免疫组化染色观察小鼠肾脏病理变化;采用Western blot和RT-PCR法观察小鼠肾脏SIRT1、氧化应激相关蛋白的变化。结果: RSV组小鼠血TG、TC、LDL-C及肌酐(CRE)水平均降低,肾组织中系膜增生和纤维化程度减少,纤维粘连蛋白(FN) 的表达增加,小鼠肾脏中SIRT1及抗氧化酶GPx4、SOD2表达增加。结论: RSV通过调控SIRT1通路提高抗氧化酶的表达,抑制肾组织氧化应激反应改善了高脂小鼠诱导的肾功能损伤。
Objective: To investigate the effect of resveratrol (RSV) on renal function in high-fat mice and its mechanism of regulating SIRT1 to inhibit oxidative stress and improving renal injury in mice. Methods: Thirty six-week-old C57BL/6J mice were selected to construct a high-fat mouse model. The mice were divided into the control group (CTRL group), the high-fat group (HL group), and the resveratrol group (RSV group), with 10 mice in each group. After 18 weeks of feeding, the blood was collected to detect the levels of blood lipid and creatinine (CRE). The pathological changes of kidney were observed by HE and immunohistochemical staining. The changes of SIRT1 and oxidative stress related proteins in mice kidney were observed by Western blot and RT-PCR. Results: The levels of blood lipid TG, TC, LDL-C and creatinine (CRE) in RSV group were decreased, the mesangial proliferation and fibrosis in renal tissue were decreased, the expression of fibronectin (FN) was increased, and the expression of SIRT1 and antioxidant enzymes GPx4 and SOD2 in mice kidney were increased. Conclusion: RSV improves the expression of antioxidant enzymes by regulating the SIRT1 pathway, and inhibits oxidative stress in renal tissue to improve renal function injury induced by high-fat mice.
