Abstract

Objective: To study the difference of skin microecology between elderly patients with atopic dermatitis (AD) and healthy elderly people in Baotou area based on 16 SrDNA Illuminate sequencing technology, so as to provide basis for the prevention and treatment of AD. Methods: A total of 15 elderly patients with atopic dermatitis (PA group) and 10 healthy elderly people (CK group) were collected from the Department of Dermatology, the First Affiliated Hospital of Baotou Medical College from January 2024 to June 2024. PA collects skin microbiome samples from the affected area of the upper limb, and CK group samples were taken from the same part of the skin. Illuminate high-throughput sequencing was performed by extracting sample DNA. Results: (1) The samples were clustered according to 97% similarity, and a total of 7825 OTU sequences were obtained, of which the phylum, class, order, family, genus and species were 59, 132, 261, 425, 1053 and 672, respectively. (2) At the level of phylum, class, order, family, genus and species, the abundance of Firmicutes and its subordinate bacteroidetes, coccus, staphylococcus and staphylococcus in PA group was significantly higher than that in CK group (P<0.05). The abundance of Proteobacteria and its related Proteobacteria, Pseudomonas, Moraxellaceae and Acinetobacter was significantly lower than that of CK group (P<0.05). At the family level, there was no statistically significant difference in species abundance between the two groups (P>0.05). (3) The results of LEfSe analysis showed that Firmicutes and its subordinate taxa ( including Bacillus, Coccobacteria, Staphylococci, and Staphylococcus ) played a key role in the difference in bacterial composition between the two groups (P<0.05). (4) There was no significant difference in Alpha diversity between PA group and CK group (P>0.05). (5) Beta diversity analysis showed that there was a significant difference between PA group and CK group (P<0.05). Conclusion: There are significant differences in the composition and diversity of skin flora between elderly AD patients and healthy elderly people, indicating that elderly AD may be related to skin microecology.
